1-2 weeks: Most standard pregnancy tests should be positive by then. Request a blood test if there are any doubts.

Any time, really.: If you have enough HCG in your blood to cause nausea, changes in appetite, or breast enlargement, it will definitely show up in the urine, enough to turn a home test positive.
