Take a preg test: Many women spot on depo.  The only way to know for certain if you are pregnant is to take a home pregnancy test.  They are cheap and available at any local pharmacy.

Take a test: Don’t guess. Take a test.  They are the same tests used in a doctors office. They can detect a pregnancy 7-10 days after conception. The best time to take one is when you miss your period.  If you doubt the result you can repeat it in 1-2 days. If you were pregnant the hormone level would rise enough for detection.

Probably not: The depo shot is very effective contraception so you are not likely to conceive for at least three months after receiving the shot.  The most common side effect of the shot is disruption of the normal cycle.  Many women have irregular spotting and bleeding but some have no bleeding at all.  The effect of the shot on the cycle can last for up to 18 months.

Irregular : Irregular periods, coming off depo, and using Plan B can all cause irregular bleeding and missed periods making it very confusing to predict ovulation and possible pregnancy.  Since most home pregnancy tests are 97% accurate 1-2 weeks after the missed period, you can surmise that they will be accurate 3-4 weeks after the episode of unprotected sex.  The other thing you can do is to give your doctor a call.  They can check a urine test which will be more accurate a little earlier than the home pregnancy test or they can do a blood test that can tell if you're pregnant 7 days after conception. Less than 1% chance: Depoprovera, if administered on time every 12 weeks, is probably the most effective hormonal birth control option  a common side effect of  depoprovera is irregular bleeding/ spotting but 75% of patients will experience no bleeding after one year.  A blood pregnancy test can be ordered to confirm the urine test. If you are still having symptoms , you may need to be worked up for other problems.

Not likely: Depo Provera (medroxyprogesterone) is notorious for weight gain with its' use along with your other symptom of "spotting".
