None!: Assuming they are clean.  Most mens briefs are cotton and can be bleached when washed and sanitized.  Women frequently ( even in my office) wear nylon, or polyester briefs, which cannot be thorughly lceaned, and are also non-absorbent.  I recommend everyone were cotton briefs for daily wear. Sexual encounters-another matter!

: Leaking semen doesn't happen, but if you are discharging mucus or cloudy fluid, you likely have an STI like gonorrhea, chlamydia, or nongonoccal urethritis. Or maybe it's just urine. Or your just over-worried, as in your other questions about prostate fluid, dusty rooms, and dog treats. Your germophobia and OCD are showing. You probably need professional counseling, if not already in such care.

Not particularly: Men have survived for thousands of generations before underwear of any sort was devised. They actually are outside the body because they function better at a cooler than core body temperature. What you wear is a comfort/personal choice. Do what the spirit moves you to do.

If you are experiencing discomfort with the movement of your scrotum during running or jogging, wear a supportive garment to limit the movement and discomfort.
