A viral culture involves taking a sample and plating it in a culture medium that allows it to grow. There are dozens of different media for viral culture, as each virus requires a different nutrient environment for it to grow. So, whenever this is suspected, many different types of culture media are "plated" to make sure the appropriate virus can be identified.
