Could having a headache off an on and a muscle spasm under right eye be a sign of a brain tumor?

In brief: Unlikely

A brain tumor would not be the first thing that comes to mind.
If this has persisted more than a few days I suggest seeing your PCP.

In brief: Details but unlikely

There are a number of different types of "brain tumors" which can cause symptoms.
While headaches can be indicative of a mass within the brain, usually there are more significant associated symptoms other than muscle spasm which would make suspicion arise.
