In brief: May be neck muscle

If it is really behind the neck, it isn't likely to be a gland.
The glands are mainly on the front side of the neck. One of the larger back of the neck muscles may be larger than the other side, or may be swollen from being over used. This muscle is called the trapezius, if you want to look it up and confirm that is where the swelling is at.
