In brief: Could be arthritis

Arthritis frequently manifests as pain with the onset of activity and then improves with continued activity, and may get worse with too much activity.
Anti-inflammatory medicines are a good place to start. Exercise and weight loss also help.

In brief: I

I recommend consulting with a rheumatologist.
There are multiple causes for knee pain that may be missed by a general practictioner. Gout, ra, pseudogout, etc... That would be your best best at getting a diagnosis and the appropriate treatment.
