In brief: Exam,imaging

Workup would involve physical examination, and appropriate imagine (at minimum an x-ray of the affected joint).
Early arthritis or degenerative joint however may not be reflected on an xray.

In brief: Complete Evaluation

Arthritis is diagnoised by first taking a complete history and doing a physical exam. If tests are needed to make the diagnosis, additional information can be obtained from laboratory tests, joint aspiration, ultrasound and x-rays.
