In brief: Ther is no best time
Perimenopause symptoms can start anytime from the 30's to the 50's.
The symptoms can be constant or intermittent. Although tests can be done, many studies have shown that they are unreliable and can change from postmenopausal levels back to premenopausal levels. Perimenopause is a diagnosis that is made from symptoms, not tests.

In brief: Difficult to tell
Perimenopause is difficult to diagnose by blood test.
The test for menopause, an fsh level, typically does not go up until the time of menopause itself. Day 2, 3 or 4 is the correct day to do an fsh level, but it will not necessarily help diagnose perimenopause though it may be help predict outcomes of fertility treatments.
