Is an ANA of 1:640 (multiple times) significant if many of the symptoms of a connective tissue disease are there but other bloodwork including specific antibodies are negative?

Diagnosis of lupus depends on presence of at least 4 criteria from a multitude of criteria not just symptoms.
Examples include arthritis, ana, low white or red blood count, pleurisy, confusion, rash, etc. Some of the criteria may occasionally have different reasons other than lupus. Your doctor can address with you what criteria you may actually have that count towards lupus.

Tests are tests. The level of ANA positivity has little or no relationship to disease activity.
A high titer ANA only means it is significant enough to follow. Tests are now so sensitive that they may precede disease by years and years. Anas can be induced by common viral illnesses, albeit transiently. So don't worry. You did not specify sx, but at this point you only need rx for your ills!
