The scrotum may be asymmetric - but the testes are usually - you can feel them if you gently roll the scrotum between your fingers - the same size.
Your son should be checked by his pediatrician - a simple screening exam and possibly an ultrasound.

Screening for testicular cancer prior to puberty is not generally recommended. New changes in size, color or position of the testicles would be cause for concern.

Testicles can appear to be different sizes due to asymmetry of the scrotum. Cancer of the testes is very very rare in a child.
Self exam should start in puberty. Teach a male teen to check testicles regularly for changes or lumps. If you are concerned about a significant difference in size see your pediatrician.
