In brief: Clinical exam
Clinical examination by a knowledgeable physician.
Tests rule out other entities, but do not diagnose fibromyalgia. Tsh is valuable, as hypothyroidism can mimic fibromyalgia.

In brief: Rule out other stuff
Fibromyalgia is a 'diagnosis of exclusion' which means that the blood tests done during the work up of fibromyalgia are to rule out other possible diagnoses, such as lupus, rheumatoid arthritis, vitamin deficiency, & others.
I often check a sed rate (esr), rheumatoid factor (rf), vitamin d and b levels, and thyroid function. In special cases i might check an ANA or other rheumatologic tests.

In brief: Palpation
? re tests to prove fibromyalgia (FM).
The only test used to "rule in" FM is a standardized palpation of several anatomical loci to observe for pain in at least 11 of 16 points. All other tests, including imaging studies & chemical & other analyses of body fluids, are to "rule out" other possible disorders explanatory for the patient's symptoms, which should include both upper & lower back pain.
