In brief: Very small firm bump
Often the first sign of a wart starting is having a very small, hardly noticeable, almost clear, firm bump on the bottom of your foot.
To notice it you would have to softly rub over it with your fingers. If treatment starts at this stage, even with an OTC product, the chances of a quick resolution are good.

In brief: Skin line disruption
Along with the other answers, there is noticible disruption of the normal skin lines of the feet when encountering plantar warts.
Callouses will have skin lines running through them, but warts will have a "stop-and-start" distribution to the skin lines.

In brief: Pain, callus
Plantar warts are on the bottom (sole) of your foot, and are often painful to squeeze and develop callus tissue around them. They will usually disrupt the skin lines.
