In brief: A number of things

There are a number of possible causes for it.
Lack of movement can cause accumulation of fluid in the leg tissues and this can make you stiff and sore. Low back trouble can do it as can nerve irritation. If you have visible swelling associated with the pain, there is the possibility that it could be a blood clot, especially if it was a several hour long drive.
