Drove 3.5 hrs wiih break after 1.5 (left friday drove home sunday) could that cause a DVT I have mild calf cramp just worried?

It could,but rare: In a 28yo without other medical issues or bleeding problem to predispose u to blood clots this would be very unusual. Additionally, that's not a very long car ride to have stasis of the blood pooled in your leg veins. U likely have muscle cramping due to dehydration, low magnesium levels, or something else.
