Amitrip or Nortrip: Both drugs are in the same class called tricyclic antidepressants and have similar pharmacologic activity. Amitriptyline is converted by the body to nortriptyline. Amitriptyline tends to cause more sedation and nortriptyline is used when a more stimulatory side effect is desired. Amitriptyline is preferred when nightime drowsiness is desirable. How does Amitriptyline work for nerve pain?

Still unknown: We used to think that amitriptyline acted on the brain at the level of the thalamus or other pain interpretive centers but recent evidence points to a peripheral action site blocking C-type pain fibers- specifically, nicotinic acetylcholine receptors carrying neuropathic pain.
