Not necessarily: It usually takes more like 10-14 days to develop a positive. However, maybe she is pregnant from last months cycle or if she had unprotected sex more than once that could also be a possibility. Always consider confirming with a blood pregnancy test as it is less likely to give you a false negative.

Probably pregnant: earlier. Home pregnancy tests turn positive 10-14 days from the time of conception.

Depends on when you: had sex. To get pregnant, you need to have sex when you are ovulating, which is about 2 weeks after you start your period. If you have sex then and conceived, you would have a pos preg test 14 days later.
