About 45 minutes: Benadryl (diphenhydramine) can make some people drowsy. The maximum effect occurs in about 45 minutes or so. However, it can result in hyperalertness in some people.

Sure: Usually Benadryl (diphenhydramine) and its generic equivalent comes in a capsule which can't be split, but you could always empty the capsule and only take half the contents. You can also try just 1 capsule or use the liquid form. 10ml of liquid contains 25mg.
